The large tropical lucinid clam Codakia orbicularis has a symbiotic relationship with intracellular, sulfide-oxidizing chemoautotrophic bacteria. The respiration strategies utilized by the symbiont were explored using integrative techniques on mechanically purified symbionts and intact clam-symbiont associations along with habitat analysis. Previous work on a related symbiont species found in the host lucinid Lucinoma aequizonata showed that the symbionts obligately used nitrate as an electron acceptor, even under oxygenated conditions. In contrast, the symbionts of C. orbicularis use oxygen as the primary electron acceptor while evidence for nitrate respiration was lacking. Citric acid (CIT, 0.3%) was assessed for its ability to reduce the pink color defect in ground, cooked (80C) turkey breast associated with nicotinamide hemochrome (NICHEME) and nitrosyl hemochrome (NITHEME). CIT incorporation in nicotinamide-treated (NIC, 1.0%) samples (CIT plus NIC) reduced (P<0.05) redness by 51% compared to the control and 63% compared to the NIC-only treatment. CIT addition in sodium nitrite-treated (NIT, 10 ppm) samples (CIT plus NIT) was similar (P>0.05) in redness to the control and reduced (P<0.05) the redness by 43% compared to the NIT- only treatment. All treatments containing citric acid lowered the pH and increased (P<0.05) cooking loss compared to samples without citric acid. At the level tested, addition of citric acid would provide poultry processors a means to eliminate or significantly reduce the pink defect. The biogenic production of hydrogen sulfide is a serious problem associated with wastewater treatment. The aim of this study was to investigate the inhibitory effect of nitrate on the dynamics of sulfate-reducing bacteria (SRB) community in a laboratory-scale wastewater reactor, originating from a denitrifying plant using activated sludge. For this purpose, denaturing gradient gel electrophoresis (DGGE) analysis targeting the dsrB (dissimilatory sulfite reductase) gene was used in combination with chemical analyses and measurement of oxidation and reduction potential (ORP). The reactors were initially dosed with 1.0 and 4.0 g/L potassium nitrate and anaerobically incubated for 490 h. Addition of 4.0 g/L nitrate to the reactor was associated with a prolonged inhibition (over 300 h, i.e., 12.5 days) of sulfate reduction and this was consistent with a rapid decrease in ORP associated with nitrate depletion. The DGGE analysis revealed that nitrate addition remarkably attenuated a distinct group of dsrB related to Desulfovibrio, whereas other dsrB groups were not influenced. Furthermore, another sulfate reduction by Syntrophobacter in the later stages of the incubation period occurred in both reactors (regardless of the nitrate concentration), suggesting that different SRB groups are associated with sulfate reduction at different stages of the wastewater treatment process.

The study is to identify the extraction of intracellular inulinase (exo- and endoinulinase) and invertase as well as optimization medium composition for maximum productions of intra- and extracellular enzymes fromAspergillus nigerATCC 20611. From two different methods for extraction of intracellular enzymes, ultrasonic method was found more effective. Response surface methodology (RSM) with a five-variable and three-level central composite design (CCD) was employed to optimize the medium composition. The effect of five main reaction parameters including sucrose, yeast extract, NaNO3, Zn+2, and Triton X-100 on the production of enzymes was analyzed. A modified quadratic model was fitted to the data with a coefficient of determination (R2) more than 0.90 for all responses. The microbial activity within activated sludge floc is a key factor in the performance of the activated sludge process. In this study, the microenvironment of activated sludge flocs from two wastewater treatment plants (Mill Creek Wastewater Treatment Plant and Muddy Creek Wastewater Treatment Plant, with aeration tank influent CODs of 60-120 and 15-35 mg/L, respectively) were studied by using microelectrodes. Due to microbial oxygen utilization, the aerobic region in the activated sludge floc was limited to the surface layer (0.1-0.2mm) of the sludge aggregate at the Mill Creek plant. The presence of an anoxic zone inside the sludge floc under aerobic conditions was confirmed in this study. When the dissolved oxygen (DO) in the bulk liquid was higher than 4.0mg/L, the anoxic zone inside the activated sludge floc disappeared, which is helpful for biodegradation. Essential roles of microalgae during the tertiary treatment of municipal wastewater have been proven. In order to avoid wash out of the biomass and also modify the quality of the treated wastewater; some techniques such as cell immobilization have been developed. To do so, in this study four samples from two species of microalgae (Chlorella vulgaris and Chlamydomonas sp.) were determined and confirmed by taxonomic identification. The samples were immobilized in calcium alginate beads. Within 10 days the amounts of nitrate (No3(-)-N) and orthophosphate (Po4(3-)-P) were measured to calculate the removal efficacy for each individual sample. Based on the standard methods, the amount of nitrate and orthophosphate were determined every 3 days within 10 days. The results indicate that immobilized microalgae are able to remove the nutrients and reduce the amount of nitrate and orthophosphate, significantly. The present study reports in detail the LD50 determination in white mice of ICR strain from iv administration of barium in the form of chloride. The LD50 value was found to be 19.2 mg Ba2+/kg. The results for nitrate and acetate were close to chloride, but in Swiss-Webster strains these values were lower by a factor of almost 2. Further, LD50 values for Mg, Ca and Sr (using chlorides in ICR-mice) were 14.4, 42.4 and 147.6 mg/kg, respectively.

The influence of extensive sedum-moss vegetated roofs on runoff water quality was studied for four full scale installations located in southern Sweden. The aim of the study was to ascertain whether the vegetated roof behaves as a sink or a source of pollutants and whether the age of a vegetated roof influences runoff quality. The runoff quality from vegetated roofs was also compared with the runoff quality from non-vegetated roofs located in study areas. The following metals and nutrients were investigated: Cd, Cr, Cu, Fe, K, Mn, Pb, Zn, NO3-N, NH4-N, Tot-N, PO4-P, and Tot-P. The results show that, with the exception of nitrogen, vegetated roofs behave as source of contaminants. While in lower concentrations than normally found in urban runoff, some metals appear in concentrations that would correspond to moderately polluted natural water. Diffuse pollution of water resources from agricultural sources is a major environmental issue in the European Union, and has been dealt with by specific legislation: the Nitrate Directive of 1991 and the Water Framework Directive of 2000. These attempts to provide a coordinated approach to solving environmental problems require methods and tools for spatial analysis and modelling on a continental scale, with river basins being used as spatial units. This paper presents a screening model (Ag-PIE), developed in a GIS environment, for the assessment of pressures from agricultural land use and the consequent impacts on surface and groundwater. Ag-PIE has been applied at the European scale (EU15), with focus on nitrogen pollution from chemical fertilisers and manure. The model adopts a multi-criteria evaluation procedure applied to spatial data layers which represent the variety of factors affecting the pollution process. The DPSIR (Driving forces, Pressures, State, Impact, Responses) approach is applied to provide the modelling approach with a conceptual framework and to further analyse and communicate results. Ag-PIE is ultimately aimed at providing a tool making use of state-of-the-art geographical databases to support policy-makers at the European level. The scale of reference adopted is the river basin, in particular those that extend across national boundaries. The quality of the results obtained has been assessed against existing related studies and monitoring reports and by means of sensitivity analysis. Conclusions are driven by considering the potential of Ag-PIE in devising policy support and its strengths and weaknesses in view of identifying future research needs.

Sulfide accumulation due to bacterial sulfate reduction is responsible for a number of serious problems in the oil industry. Among the strategies to control the activity of sulfate -reducing bacteria ( SRB ) is the use of nitrate, which can exhibit a variety of effects. We investigated the relevance of this approach to souring oil fields in Oklahoma and Alberta in which water flooding is used to enhance oil recovery. SRB and nitrate -reducing bacteria ( NRB ) were enumerated in produced waters from both oil fields. In the Oklahoma field, the rates of sulfate reduction ranged from 0.05 to 0.16 M S day À 1 at the wellheads, and an order of magnitude higher at the oil -water separator. Sulfide production was greatest in the water storage tanks in the Alberta field. Microbial counts alone did not accurately reflect the potential for microbial activities.
